Protonation studies and multivariate curve resolution on oligodeoxynucleotides carrying the mutagenic base 2-aminopurine.
2-Aminopurine (P) is a mutagen causing A.T to G.C transitions in prokaryotic systems. To study the base-pairing schemes between P and cytosine (C) or thymine (T), two self-complementary dodecamers containing P paired with either C or T were synthesized, and their protonation equilibria were studied by acid-base titrations and melting experiments. The mismatches were incorporated into the self-c...
متن کاملSynthesis and Properties of Oligodeoxynucleotides Carrying 2-Aminopurine
The use of benzoyl, isobutyryl and dimethylaminomethylidene groups for the protection of the exocyclic amino function of 2-aminopurine during oligonucleotide synthesis has been investigated. Best results in the synthesis were obtained with the monomers of 2-aminopurine protected with the isobutyryl group.
متن کاملQuantum chemical characterization of the cytosine: 2-Aminopurine base pair
The nature of the base pairing between cytosine and 2-aminopurine is investigated by means of quantum mechanical calculations including electron correlation and accounting for the effects of aqueous solvation. At neutral pH, both a neutral wobble base pair and a Watson–Crick-like base pair having a protonated 2-aminopurine are predicted to be close to one another in energy; other previously pro...
متن کاملSynthesis and characterization of oligodeoxynucleotides containing the mutagenic base analogue 4-O-ethylthymine.
A method for the preparation of oligonucleotides containing the mutagenic base 4-O-ethylthymine is described for the first time. Use of p-nitrophenylethyl type base protecting groups together with phosphitetriester solid-phase methodology makes possible the rapid and efficient preparation of oligonucleotides bearing 4-O-ethylthymine, while standard base protecting groups are not compatible with...
متن کامل2-Aminopurine fluorescence quenching and lifetimes: role of base stacking.
2-Aminopurine (2AP) is a fluorescent analog of guanosine and adenosine and has been used to probe nucleic acid structure and dynamics. Its spectral features in nucleic acids have been interpreted phenomenologically, in the absence of a rigorous electronic description of the context-dependence of 2AP fluorescence. Now, by using time-dependent density functional theory, we describe the excited-st...
